My 250GB FreeAgent was initially used for a PC. I need to use it for a Macbook now. Is it possible to reformat and use with Time Machine?

It's easy - you can just use the MacOS Disk Utility to reformat it. Use Mac Extended (Journaled). Then it'll be ready to go.
